In a significant breakthrough for reproductive medicine and genetic disease prevention, eight healthy babies have been born in the UK using a controversial yet promising technique known as three-person IVF or mitochondrial donation. This method, which has been legally approved and practiced in the UK under strict regulation, has successfully prevented the transmission of severe, often fatal mitochondrial metabolic disorders from mother to child.

The technique involves transferring the pronuclei (containing the parents’ nuclear DNA) from a fertilized egg with faulty mitochondria into a donor egg that has had its nucleus removed but still retains healthy mitochondria. This creates an embryo with parental nuclear DNA but donor mitochondrial DNA, which are responsible for energy production in cells.

Out of 22 women considered at high risk of transmitting mitochondrial disease, 8 healthy infants have been born with either no or very low levels of mutated mitochondrial DNA detected in their blood. All children are thriving and meeting developmental goals, with only one child exhibiting treatable conditions unrelated directly to mitochondrial DNA mutation.

This approach targets inherited metabolic diseases caused by pathogenic mitochondrial DNA variants, which can lead to multisystem organ failure, seizures, muscle atrophy, and early death. It addresses cases where current preimplantation genetic testing cannot prevent disease transmission because all eggs carry the mutation.

The procedure is legally approved and practiced in the UK but is banned in some countries like the US. It represents a significant advance in reproductive medicine and genetic disease prevention, offering new hope to families affected by mitochondrial disorders.

A study detailing the three-person IVF technique was published in The New England Journal of Medicine. Dr. Robert McFarland, a pediatric neurologist at Newcastle University, expressed "cautious optimism" about the results. He stated, "To see babies born without mitochondrial disease is amazing."

Dr. Zev Williams, director of Columbia University Fertility Center, considers this research an important milestone. He believes that expanding the range of reproductive options will empower more couples to pursue safe and healthy pregnancies.

One child born through this method developed hyperlipidemia (high cholesterol) and cardiac arrhythmia (irregular heartbeat), but both conditions responded to treatment. It's important to note that these conditions are not directly related to the three-person IVF technique.

However, a separate study has linked a sperm donor's genetic mutation to cancer in 10 conceived children, but this is not connected to the three-person IVF technique discussed in this article.
